Abstract:In order to isolate andscreen microorganisms thatcould degrade ligninefficiently
and then constructbiological immobilizationcarrier
there was 1 strain of high effcient lignindegrading fungi
named G13
isolated from the sludge samples collected by the paper mill
which the highestlingin enzyme activity of manganese peroxidase(MnP)
laccase(Lac) and ligninperoxidase(LiP) were 8.27
0.95and 13.55U/L
respectively. Throughthe study of factors that can affect the pelletization
the best conditions areas follows: sucrose concentration of 18g/L and ammonium tartrate concentrationof 2.5g/L wereworked as the best carbon source and nitrogen source
respectively; the mostappropriate medium pH was 5; 5mL of spore suspension was inoculated on 100mL ofliquid medium forpelletization (in 250mL Erlenmeyer flask);the optimumtemperature was 30℃; and the most suitableshaker speed was 160r/min. Cultured under the most suitable growing conditions for 72h
themycelium pellet formed by Aspergillus fumigatus G13 reached its maximum diameterof 3.4mm withsmooth surface
excellent flexibility and hollow structure
which can be usedas biological immobilization carrier as wellas meets the needs of practicalapplication such as industrial wastewater treatment.
